Peptides for Kidney Function and Kidney Health: Klotho, SS-31, MOTS-c, Humanin & Vilon

Can peptides support kidney function and renal health? Emerging research is exploring peptides that influence mitochondrial function, oxidative stress, inflammation, fibrosis and cellular aging.

Among the most interesting molecules are Klotho, SS-31, MOTS-c, Humanin, BPC-157, ARA-290 and Vilon.

Klotho: A Key Peptide for Kidney Aging

Klotho is a protein produced predominantly by the kidney and plays an important role in phosphate metabolism, FGF23 signaling, mineral balance and renal protection.

Klotho levels tend to decline during chronic kidney disease, and lower circulating Klotho has been associated with reduced eGFR and poorer renal outcomes. A meta-analysis of 8 cohort studies involving 3,586 people found that soluble Klotho was positively correlated with eGFR, while lower levels were associated with increased adverse kidney outcomes.

Experimental research is investigating Klotho as a potential target for renal fibrosis, inflammation, oxidative stress and kidney aging. Recent work continues to refine its role in CKD and mineral metabolism.

SS-31: Targeting Mitochondrial Kidney Health

SS-31, or Elamipretide, is a mitochondria-targeted peptide.

Because kidney tubular cells have high energy requirements, mitochondrial dysfunction can contribute to cellular injury. SS-31 is being investigated for its ability to influence mitochondrial function and oxidative stress, including in experimental models of kidney injury.

This makes SS-31 an important research peptide for mitochondrial renal protection.

MOTS-c: Metabolic and Mitochondrial Signaling

MOTS-c is a mitochondrial-derived peptide involved in metabolic regulation and cellular stress responses.

Researchers are investigating its relationship with AMPK signaling, glucose metabolism, mitochondrial function and aging. Studies in people with advanced kidney disease have also examined circulating MOTS-c, although these findings do not establish MOTS-c as a kidney treatment.

Humanin: Cellular Protection

Humanin is another mitochondrial-derived peptide being studied for its effects on cellular survival, oxidative stress and inflammation.

Experimental research involving Humanin analogues has reported protective effects in models of kidney injury, making it a promising area for future investigation into mitochondrial and cellular renal protection.

BPC-157 and ARA-290

BPC-157 has been investigated experimentally for tissue repair, endothelial function, inflammation and oxidative stress, including models involving renal injury.

ARA-290, also known as cibinetide, is an erythropoietin-derived tissue-protective peptide being investigated for anti-inflammatory and cytoprotective effects.

Both remain investigational for kidney applications.

What About Vilon?

Vilon is a short thymic peptide studied primarily in the context of immune and cellular regulation and longevity research.

Compared with Klotho or mitochondrial peptides such as SS-31, there is considerably less kidney-specific evidence for Vilon. It should therefore be regarded as an experimental longevity peptide rather than an established renal therapy.

Klotho is particularly interesting because it sits at the intersection of kidney function, mineral metabolism and aging, while mitochondrial peptides such as SS-31, MOTS-c and Humanin represent another promising research direction.

Many of these peptides remain investigational, and evidence from laboratory or animal studies should not be interpreted as proof of clinical efficacy in humans.
